Fast decellularization process using supercritical carbon dioxide for trabecular bone

Decellularization is a process that consists on the removal of immunogenic cellular material from tissue, so it can be safely implanted as functional and bioactive scaffold. Most decellularization protocols rely use harsh chemicals very long washing processes, leading to severe changes in ultrastructure loss mechanical integrity. To tackle these challenges, supercritical carbon dioxide (scCO2) herein proposed an alternative methodology for assisting porcine trabecular bone tissue combined, first time, with Tri(n-butyl) phosphate (TnBP). Histological DNA analysis revealed both TnBP scCO2 were able extract content scaffolds, being this effect more pronounced treatments used co-solvent. The combined protocol led decrease by at least 90%, demonstrating potential opening new possibilities future optimizations.
